Öğe Antimicrobial susceptibility, presence of resistance genes and biofilm formation in coagulase negative staphlococci isolated from subclinical sheep mastitis(Veteriner Fakultesi Dergisi, 2012) Ergün, Yaşar; Aslantaş, Özkan; Kireçci, Ekrem; Öztürk, Fatma; Ceylan, Ahmet; Boyar, YilmazIn this study, coagulase negative staphylococci (CoNS) (n = 70) isolated from subclinical sheep mastitis were screened for minimum inhibitory concentration (MIC) to antimicrobials used commonly in veterinary field in Turkey. In addition, plasmid profiling and biofilm production of CoNS isolates was investigated. All isolates were found to be susceptible to amoxycillin-clavulanic acid, cephalothin, gentamicin, enrofloxacin and oxacillin. The highest resistance was observed in 42.9% (n = 30) of the isolates against the beta-lactam antibiotics, penicillin and ampicillin. All beta-lactam resistant isolates produced beta-lactamase and carried blaZ. Tetracycline resistance was observed in 11.4% (n = 8) of the isolates, either alone or in combination with beta-lactams and macrolides. Of the tetracycline resistant 8 isolates, 5 carried the tetK gene, one carried the tetM and 2 isolates carried both genes together. Erythromycin resistance was observed in 5.7% of the isolates; msrA was detected alone (one isolate) or in combination with mphC (one isolate) and ermC (one isolate). ermA was observed only in one isolate. Most of the strains showed only a single plasmid band in size of 19.3 kb, but some had 2 to 3 plasmids ranging from >19.3 kb to 0.9 kb. Out of 70 CoNS isolates, 28 (40%) were identified as biofilm producer by Congo red agar (CRA) method, and 30 (42.9%) were positive for both icaA and icaD genes, which are known to be responsible for biofilm formation in CoNS.Öğe Antimicrobial susceptibility, presence of resistance genes and biofilm formation in coagulase negative staphlococci ısolated from subclinical sheep mastitis(2012) Ergün, Yaşar; Öztürk, Fatma; Aslantaş, Özkan; Ceylan, Ahmet; Kireççi, Ekrem; Boyar, YılmazBu çalışmada, subklinik koyun mastitislerinden izole edilen koagulaz negatif stafilokokların (KNS) (n = 70) veteriner hekimlik alanında yaygın olarak kullanılan antimikrobiyallere minimal inhibitor konsantrasyonları (MİK), plazmid profilleri ve biyofilm üretimi araştırıldı. Tüm izolatlar amoksisilin-klavulanik asid, cefalotin, gentamisin, enrofloksasin ve oksasiline duyarlı bulundu. En yüksek direnç izolatların %42.9’unda (n = 30) beta-laktam antibiyotiklere (penisilin ve ampisilin) karşı gözlendi. Beta-laktam antibiyotiklere dirençli izolatların tamamı beta-laktamaz sentezlediği ve blaZ genini taşıdığı belirlendi. Tetrasiklin direnci tek veya beta-laktam antibiyotikler ve makrolidlerle kombine olarak izolatların %11.4’ünde (n = 8) gözlendi. Tetrasiklin dirençli izolatların 5’inde tetK, birinde tetM ve ikisinde de her iki geni birlikte bulundu. Eritromisin direnci izolatların %5.7’sinde (n = 4) bulundu. msrA, mphC, ermA birer izolatta, ermC ise msrA geni ile kombine olarak bir izolatta tespit edildi. İzolatların büyük kısmı 19.3 kb büyüklüğünde tek plasmid gösterirken, bazı izolatlar büyüklüğü >19.3 kb ile 0.9 kb arasında değişen 2-3 plazmid taşıdığı belirlendi. Yetmiş KNS izolatından 28’i (%40) Kongo Red Agar (KRA) metodu ile biyofilm sentezi ve 30’unun da (%42.9) biyofilm sentezinden sorumlu olan icaA ve icaD genleri yönünden pozitif olduğu saptandı.Öğe Change in the dimensions of the lumbar area muscles after surgery: MRI analysis(Kare Yayıncılık, 2020) Duman, Fatma; Serarslan, Yurdal; Öztürk, Fatma; Yücekaya, Bircan; Atçı, NesrinObjective: This study aims to assess the change in the dimensions of the lumbar muscles in patients with chronic lower back pain using Magnetic Resonance Imaging (MRI) and to determine pre/post effects of surgery. Methods: We enrolled 28 individuals (13F/15M; age: 45.39±11.56 years) whose L2-S1 muscle measurements were obtained using MRI, before and at follow-up 6-12 months after surgery. The control group comprising 37 individuals (18F/19M; age: 34.41±10.72 years) who had no lumbar pathology but for whom retrospective archive images were available. In the axial MRI analysis, the cross-sections of m.multifidus, mm.erector spinae and m.psoas major on both sides were measured with the 'closed polygon' technique. Results: The L2-3 and L4-5 levels of the m.multifidus on the right side, the L2-3, L4-5 and L5-S1 levels of the m.multifidus and the L5-S1 levels of the mm. erector spinae on the left side cross-sectional areas were significantly lower than the control group (p<0.05). The right-side m.multifidus and the left-side mm.erector spinae sectional areas were significantly lower than the pre-surgery values at the L5-S1 levels (p<0.05). Conclusion: This study demonstrated that chronic lower back pain causes atrophy in the lumbar muscles and established the existence and continuity of atrophy after surgery.Öğe Characterization of Staphylococcus aureus strains isolated from subclinic bovine mastitis by protein patterns, antibiotic resistance and plasmid profile(2006) Aslantaş, Özkan; Öztürk, Fatma; Çelebi, Ayten; Açık, Leyla; Ergün, YaşarHatay bölgesindeki subklinik inek mastitislerinden izole edilen toplam 50 Staphylococcus aureus susunun protein paterni, antibiyotik dirençliliği ve plazmid profili yönünden karakterizasyonu yapıldı. S. aureus suşlarının SDS-PAGE ile analizinde moleküler ağırlığı <14.4->116 kDa arasında değişen 17-49 bant elde edildi. Elektroforetik protein bantlarının bilgisayar destekli numerikal analizi sonucunda S. aureus suşları 2 gruba ayrıldı. S. aureus suşları trimethoprim-sülfametoksazol, danofloksasin, amoksisilin-klavulanik asit, sulbaktam-ampisilin, eritromisin, linkomisin, sefoperazon, oksitetrasiklin, amoksisilin, streptomisin ve penisilin'e sırasıyla % 4, % 4, % 10, % 14, % 30, % 34, % 36, % 38, % 50, % 54 ve % 68 oranlarında dirençli bulundu. İncelenen izolatların 47'sinde, büyüklüğü 1.7->24 kb arasında değişen plazmidler saptandı.Öğe Numerical analysis of Candida species from uriner system infections based on SDS-PAGE and detection of antifungal resistance(TIP ARASTIRMALARI DERNEGI, 2007) Duran, Nizami; Öztürk, Fatma; Açik, Leyla; Aslantaş, Özkan; Aslan, GönülAim: The aim of the present study was to evaluate the protein patterns and numerical analysis of Candida species isolates from urinARY system infections by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE), and to detect antifungal susceptibility. Methods: In this study, 96 Candida spp. belonging to 46 strains of C.albicans, 18 strains of C. glabrata, 14 strains of C. tropicalis, 8 strains of C. krusei, 6 strains of C. parapsilosis, 3 strains of C. dubliensis and 1 strain of C. kefyr from urine samples or urinary system infections were examined by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The susceptibility of Candida strains against antifungal agents were assessed by minimal inhibitory concentrations (MIC). Results: Nineteen distinct band patterns in the range of 63-120 kDa were obtained. Different isolates of each species were clearly different in each of the seven species. But, some variation in the protein patterns was detected within the same species. Conclusion: The whole-cell protein profiles performed by SDS-PAGE associated with computer-assisted numerical analysis may provide preliminary criteria for taxonomic and epidemiological studies of such microorganisms.
