Öğe Effects of ebselen on radiocontrast media-induced hepatotoxicity in rats(Sage Publications Inc, 2013) Basarslan, Fatmagul; Yilmaz, Nigar; Davarci, Isil; Akin, Mustafa; Ozgur, Mustafa; Yilmaz, Cahide; Ulutas, Kemal TurkerOxidative stress is accepted as a potential responsible mechanism in the pathogenesis of radiocontrast media (RCM)-induced hepatotoxicity. Therefore, we aimed to investigate the protective effects of ebselen against RCM-induced hepatotoxicity by measuring tissue oxidant/antioxidant parameters and histological changes in rats. Wistar albino rats were randomly separated into four groups consisting of eight rats per group. Normal saline was given to the rats in control group (group 1). RCM was given to the rats in group 2, and both RCM and ebselen were given to the rats in group 3. Only ebselen was given to the rats in group 4. Liver sections of the killed animals were analyzed to measure the levels of malondialdehyde (MDA) and activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px), as well as histopathological changes. In RCM group, SOD and CAT levels were found increased. In RCM-ebselen group, MDA, SOD and CAT levels were found decreased. In RCM-ebselen group, however, GSH-Px activities of liver tissue increased. All these results indicated that ebselen produced a protective mechanism against RCM-induced hepatotoxicity and took part in oxidative stress.Öğe Investigation of antiseptic resistance genes in Staphylococcus spp. isolates(Natural and Engineering Science, 2020) Akin, Mustafa; Özcan, Birgül; Cantekin, Zafer; Ergün, Yaşar; Bulanık, DilşadAntiseptic and disinfectants are used very frequently in all health institutions, including hospital and veterinary application areas, in the home environment and food production industry, to prevent infections and contaminations. At present, the quaternary ammonium compounds, benzalkonium chloride and chlorhexidine digluconate is one of the divalent cations most commonly used chemicals such as antiseptics and disinfectants. However, the widespread use of biocides has brought about the emergence of bacteria resistant to antiseptics/disinfectants. It is known that bacteria develop resistance mechanisms against antibiotics as well as disinfectants. Epidemiological data on antiseptic susceptibility and distribution of resistance genes are very important for nosocomial infections. Some species, including the species belong to genus Staphylococcus, cause foodborne poisoning and various clinical infections such as skin and soft tissue and surgical site infections, endocarditis, mastitis, pneumonia and bacteremia in humans and animals. Staphylococcus strains can contain plasmid-derived qacA/B and qacC genes that provide resistance to quaternary ammonium compounds (QAC). In this study, the presence of antiseptic resistance genes (qacA/B and qacC) in 90 Staphylococcus spp. strains isolated from chicken carcass, bovine tank milk, various cheeses and bovine clinical mastitis samples were determined by simplex polymerase chain reaction. QacA/B was found in %18.8 and qacC in %2.2 of the studied isolates. Of antiseptic resistance genes, qacA/B was detected in cheese and bovine clinical mastitis samples, and qacC in chicken carcass. © 2020, Natural and Engineering Science. A 17-year-old female patient was admitted to the department of chest diseases with respiratory symptoms. The patient had undergone a thyroid surgery for goiter at the age of 9. A bilateral nodular opacity was detected by radiological examination. The histopathologic examination of the specimen obtained from computed tomography guided trucut biopsy was diagnosed as PTC. We present a very rare case of PTC with lung metastasis that had undergone subtotal thyroidectomy due to dyshormonogenetic goiter eight years ago.Öğe Tumor budding and E-Cadherin expression in endometrial carcinoma: Are they prognostic factors in endometrial cancer?(Academic Press Inc Elsevier Science, 2012) Koyuncuoglu, Meral; Okyay, Emre; Saatli, Bahadir; Olgan, Safak; Akin, Mustafa; Saygili, UgurObjective. To evaluate the prognostic value of tumor budding (TB) in endometrioid (EEC) and non-endometrioid endometrial cancers (NEEC) and to determine its correlation with expression of E-cadherin. Methods. Ninety-five patients with primary endometrial carcinoma were examined statistically. All patients were diagnosed, treated, and given follow-up care at Dokuz Eylul University Faculty of Medicine. Tumor budding detected by either H&E-stained sections and anticytokeratin-staining C11. The tissue block with the largest invasive front was chosen for budding counting and immunostaining. E-cadherin expression was examined by immunohistochemistry using the primary antibodies against to it. Results. Tumor budding was low-grade in 73 and high-grade in 22 cases. E-cadherin expression loss was identified in 48 patients. The high-grade TB was significantly higher in patients with advanced stage and deep myometrial invasion (p = 0.032 and 0.018, respectively). E-Cadherin expression was significantly lower in NEECs than EECs (p=0.032). The negative expression of E-cadherin was associated with advanced stage and poor differentiation (p = 0.001 and p = 0.024, respectively). We determined that tumor budding adversely correlated with the presence of E-cadherin expression but not statistically significant. Based on the results of multivariate analysis, TB has an independent impact on cumulative overall survival. We found no statistically significant difference between E-cadherin expression and survival. Conclusions. TB is associated with undifferentiated tumor, advanced stage and decreased postoperative survival in endometrial cancer. It might be a valuable prognostic clinicopathologic factor which can be applicable in routine examination. (C) 2011 Elsevier Inc. All rights reserved.
