    Antioxidant activities and fatty acid composition of wild grown myrtle (Myrtus communis L.) fruits

    The fruits of eight myrtles, Myrtus communis L. accessions from the Mediterranean region of Turkey were evaluated for their antioxidant activities and fatty acid contents. The antioxidant activities of the fruit extracts were determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and beta-carotene-linoleic acid assays. The fatty acid contents of fruits were determined by using gas chromatography. The methanol extracts of fruits exhibited a high level of free radical scavenging activity. There was a wide range (74.51-91.65%) of antioxidant activity among the accessions in the beta-carotene-linoleic acid assay. The amount of total phenolics (TP) was determined to be between 44.41-74.44 mu g Gallic acid equivalent (GAE)/mg, on a dry weight basis. Oleic acid was the dominant fatty acid (67.07%), followed by palmitic (10.24%), and stearic acid (8.19%), respectively. These results suggest the future utilization of myrtle fruit extracts as food additives or in chemoprevention studies.

    Chemical composition, antioxidant activities and total phenolic content of Arbutus andrachne L. (Fam. Ericaceae) (the Greek strawberry tree) fruits from Turkey

    Arbutus andrachne L and A. unedo L are two members of the Ericaceae family. The species are distributed in coastal parts of Anatolia and have edible fruits. We determined several characteristics of eight A. andrachne accessions collected from different parts of the Mediterranean regions of Anatolia. A. unedo is also represented with a sample. Notable differences were found among A. andrachne accessions. On average, the fruit width and length were 12.3 and 10.8 mm, respectively. All of the element concentrations that were tested varied considerably. The average of the total organic acids was 0.47 g/100 g fruit. Among the organic acids measured, malic acid had the highest content (0.34 g/100 g fruit) followed by ascorbic (0.10 g/100 g fruit) and citric acids (0.03 g/100 g fruit). The average for total sugars of the A. andrachne accessions was 16.38 g/100 g fruit. Fructose was the main sugar (average of 9.75 g/100 g fruit) followed by glucose (5.33 g/100 g fruit) and sucrose (1.19 g/100 g fruit). Among the A. andrachne accessions tested, the total phenolics ranged from 2422 to 4102 mu g gallic acid/g fresh weight (fw) with an average of 3343 mu g gallic acid equiv. g/fw. Antioxidant activities, determined by TEAC (Trolox equivalent antioxidant capacity) and FRAP (Ferric reducing ability of plasma), reached up to 36.1 and 29.5 mu mol Trolox equiv. (TE) g/fresh weight, respectively. It can thus be concluded that A. andrachne is a good source of phenols and antioxidants. (C) 2010 Elsevier Inc. All rights reserved.

    Effects of Bio-Bor Fertilizer Applications on Fruit Yield, Antioxidant Enzyme Activity and Freeze Injury of Strawberry

    Deficiency of Boron (B) is widespread in the many parts of region of Turkey. So, the effects of boron and plant growth promoting bacteria (Bio-B) on the fruit yield, antioxidant enzyme activity and plant freeze injury of strawberry cv. Fern were investigated under field conditions between 2013 and 2014. The experimental plot was a completely randomized design with 4 replicates. Control and Bio-B were used as fertilizer agent in the experiment. Bio-B fertilizer was applied in three methods as soil, foliar and soil + foliar application methods to strawberry plants. Data through 2 years showed that the use of Bio-B significantly increased fruit yield, antioxidant enzyme activity and decreased freeze injury of strawberry leaf. Soil + foliar applications of Bio-B fertilizer increased to fruit yield compared to the control by 55.91 %. However, foliar application of Bio-B fertilizer increased to catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) enzyme activity compared with the control treatment 41.86 %, 48.99 %, and 26.59 %, respectively and decreased freeze injury of strawberry leaves 27.41 %. Overall, the results of this study suggest that Bio-B fertilizer application have the potential to increase the yield, antioxidant enzyme activity and decreased freeze injury of strawberry plants under field conditions.

    Molecular characterization of mulberry accessions in turkey by AFLP markers

    Mulberries (Morus L.) show a great deal of genetic variability and adaptability to various environments. There are more than 24 species of mulberries in cultivated and wild forms. In Turkey, three Morus species, M. alba L., M. nigra L., and M. rubra L., are grown. In this study, we attempted to characterize 43 Morus accessions originating from distinct regions of Turkey using fluorescent dye amplified fragment length polymorphism (AFLP) markers and capillary electrophoresis. The accessions belonged to M. alba, M. nigra, and M. rubra; M. alba consisted of white- and purple-fruited samples. Eight primer combinations generated a total of 416 bands, 337 of which were polymorphic (80.5%). Resolving powers of the AFLP primers ranged from 0.410 to 0.942 making a total of 5.015, whereas the polymorphic information content ranged from 0.662 to 0.898 with an average of 0.812. Unweighted pair-group method of arithmetic mean (UPGMA) clustering of the accessions showed three major groups representing M. nigra, M. rubra, and M. alba accessions. The M. alba group had two subgroups that were not correlated with fruit color. The UPGMA dendrogram of average taxonomic differences confirmed these results. The principle coordinate analysis demonstrated that M. nigra accessions had limited genetic variation. In conclusion, our study indicated that M. nigra and M. rubra are molecularly distinct from M. alba. Our results also suggest that M. nigra accessions having a low level of morphological variation are molecularly similar.

    MOLECULAR CHARACTERIZATION OF SOME SELECTED PERSIMMON GENOTYPES AND CULTIVARS BY SRAP AND SSR MARKERS

    In this study, SRAP and SSR markers were employed to determine genetic relationships among 42 persimmon genotypes (Diospyros kaki Thunb) obtained from Hatay province and 3 persimmon cultivars, 2 of which belong to Diospyros kaki Thunb and one belongs to Diospyros oleifera Cheng. Genetic relationships were determined by using a total of 29 molecular DNA primers (SRAP and SSR). Of these primers, 21 SRAP primer combinations produced a total of 107 bands and 77.6% of them were polymorphic; 8 SSR primers produced 26 polymorphic bands with an average polymorphism ratio of 84.6%. The SRAP and SSR markers produced 4.6 bands as average and the number of bands produced per marker was calculated as 3.6. The lowest similarity was observed between MK-113 (Diospyros oleifera Cheng) and the other genotypes all belongs to Diospyros kaki Thunb (with similarity ratios of 0.41-0.69 for SRAP primers, between 0.25-0.67 for SSR primers). The genotypes/cultivars belongs to Diospyros kaki had similarity ratio between 0.98-1.00 according to SRAP and SSR markers. This synonym or similarity could be results of clonal propagation rather than autogamy.

    Phenotypic variation in physico-chemical properties among cactus pear fruits (Opuntia ficus-indica (L.) Miller) from Turkey

    Several physico-chemical properties such as fruit weight and dimensions, skin width, external and internal fruit color, vitamin A, vitamin C, acidity, pH, soluble solid content, antioxidant activity and total phenolic content of twenty five selected promising cactus pear accessions were determined. Antioxidant activity and total phenolic content of fruits were determined by -Caroten bleaching and Folin-Ciocalteu assays. The results showed great qualitative and quantitative differences in the physico-chemical characteristics of cactus pear accessions. Vitamin A (beta carotene) and vitamin C (ascorbic acid) content of genotypes were found between 2.64 and 25.13 (g/g) and 18.04 and 37.31 mg/100 g. Antioxidant activity and total phenolic content of cactus pear genotypes were between 45.5-76.8 and 19.4-49.4 mg gallic acid equivalent per g dry weight basis. The results provide important information on how to make the best use of cactus pear genotypes investigated for different uses, which is of significance for both technological research and processing practice.

    Phytochemical characterization of several hawthorn (Crataegus spp.) species sampled from the Eastern Mediterranean region of Turkey

    Background: We evaluated the total phenolic content, antioxidant capacity as well as antioxidant activity of five Crataegus species (A1, A2, Y1, Y2, Y4 accessions of Crataegus aronia var. aronia; B2, B3, B5, B6, B7, B9, Y5 accessions of C. aronia var. dentata; B10 accession of C. aronia var. minuta; Y3 accession of Crataegus orientalis var. orientalis and A3 accession of Crataegus monogyna subsp. azarella). Materials and Methods: Antioxidant activity and total phenolic content of fruits were determined by -carotene bleaching and Folin-Ciocalteu assays. Antioxidant capacity was determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Results: C. monogyna subsp. azarella had the highest total phenol, antioxidant activity and antioxidant capacity of 55.2 mg gallic acid equivalents (GAE)/g dry weight (DW), 81.9 and 31.2, respectively. C. aronia var. aronia was found to have the lowest total phenolic content (35.7 mg GAE/g DW). The antioxidant activities of fruit extracts increased in the order of C. orientalis var. orientalis < C. aronia var. minuta < C. aronia var. dentata < C. aronia var. aronia < C. monogyna subsp. azarella according to -carotene/linoleic acid assay. In recent years, C. aronia var. dentata has gained importance as a commercial species in this region. B3 and B7 accessions had fruit weight more than 14 g, and considerable total phenol content, antioxidant activity and antioxidant capacity. Conclusion: This investigation shows the potential value of hawthorn fruit species as a good source of natural antioxidants and that consumption of hawthorn fruit or its products may contribute substantial amounts of antioxidants to the diet.

    Relationships among myrtle accessions from Turkey as revealed by fruit characteristics and RAPD

    Myrtle, Myrtus communis L., is one of the most important and widespread shrubs (maquis) throughout the Mediterranean region. Its fruits ripen during the period of mid-fall to early winter and are generally dark-colored although while-colored wild forms rarely exist. In Turkey, both forms are present in addition to a while and large-fruited type, cultivated in small orchards and marketed locally. We studied two sets of myrtle accessions from Hatay, Turkey. Set I consisted of two wild accessions, dark- and white-colored forms, and six cultivated selections, while Set 2 contained three open-pollinated individuals from each accession of Set 1. We evaluated fruit characteristics of including fruit weight, width, length, soluble solids, pH and acidity. The accessions considerably differed in all the traits tested except for soluble solids. We also examined genotypic variation of Sets I and 2 using 26 RAPD primers and analyzed the data by cluster and principle components analyses. The results revealed that although some of the half-sib families (e.g., 8 a, b and c) were grouped based on their maternal parents, no overall pattern was apparent. These results suggest that myrtle pollination appeared to be not restricted to the self-pollination and the cultivated types have a narrow genetic base.

    Some selected physico-chemical characteristics of wild and cultivated blackberry fruits (Rubus fruticosus L.) from Turkey

    In this study, some selected physico-chemical properties such as fruit weight, fruit length, fruit width, total soluble solids (TSS), titratable acidity, TSS/acidity ratio, pH, total phenolic content, antioxidant activity and free radical scavenging capacity of 9 cultivated and 16 selected wild blackberry (Rubus fruticosus L.) genotypes grown in Turkey were investigated. The total phenolic content, antioxidant activity and free radical-scavenging capacity of blackberry cultivars and genotypes were determined by using Folin-Ciocalteu, beta-carotene bleaching and DPPH radical assays. The results showed that, average fruit weight and fruit dimensions were higher in cultivated blackberries than wild materials. However, TSS, acidity and pH values were higher in wild materials. The total phenolic contents of blackberry cultivars and wild genotypes were in a range of 584 (cv. Bartin) to 788 (cv. Chester) mg/100 g and 610 (Genotypr R2) to 1455 mg/100 g (Genotype R16), expressed as gallic acid equivalents (GAE), on afresh weight basis. Antioxidant activity of cultivated and wild growing blackberry fruits was found between 72.15 (cv. Arapaho)-89.75% (cv. Bursa 3) and 59.85 (R1)-87.42% (R10), respectively. The antioxidant activity of standard BHA was 85.07%. Different cultivars grown in same location consistently showed differences in antioxidant capacity. The results of this study outlines that the blackberry fruits tested are good sources of natural antioxidants.

    Total phenolics and antioxidant activity of jujube (Zizyphus jujube Mill.) genotypes selected from Turkey

    We determined the total phenolic content and antioxidant activity of methanol extracts from fifteen selected jujube genotypes endogenous to the Mediterranean region of Turkey. Total phenolic content of the fruits was analyzed by Folin-Ciocalteu colorimetric method, while the total antioxidant activity was analyzed using the beta-carotene bleaching, ferric ion reducing antioxidant power (FRAP) and 2,2-diphenil-1-picrylhydrazyl radical scavenging capacity (DPPH) assays. The highest total phenolic content was observed in MHS 6 and MHS 7 genotypes (42 and 40 mg gallic acid equivalent (GAE) g(-1) dry weight (DW)), while the lowest content was found in MHS 5 and MHS 14 (28 and 25 mg GAE g(-1) DW). MHS 13 was among the genotypes with the highest antioxidant capacity in all three methods tested (1237 mu mol g(-1) in FRAP, 83% in beta-carotene bleaching method and 99% in DPPH). The present study demonstrates the potential value of jujube genotypes for pharmaceuticals and nutrition.
