K.; Urer, Koldas E.; Naseer, Z.; Dogruer, G.; Karaca, F.; Cetin, Coskun N.Background: Periostin (POSTN) is an extracellular matrix (ECM) protein that plays an important role in the metastatic process and cancer cell migration. As implantation is a similar mechanism to metastasis, it has been hypothesized that POSTN may also play a role in the implantation process. Aims: The aim of the present study was to compare POSTN and progesterone levels during the early pregnancy stage in Damascus goats. Methods: Forty goats were synchronized using progesterone based sponges and were mated upon estrus signs display. While ten goats were kept as control (CON) and were not allowed to mate. Blood samples were taken through jugular venepuncture from CON and synchronized goats on day 13, 15, 17, 19, and 21 of breeding. Progesterone and POSTN levels were determined by enzyme-linked immunosorbent assay (ELISA). Later the pregnancy diagnosis was confirmed by transabdominal ultrasonography on day 50 after mating. Results: Progesterone level was influenced by status of pregnancy and day of observation with an interaction between the status of pregnancy and day of observation in goats. Whereas POSTN level was only affected by the day of observation. Conclusion: POSTN level did not vary with progesterone level during phase of embryonic implantation in goats; however, standardization and application of different procedures for POSTN assay in a large group of animals might be useful as an early pregnancy biomarker in goats.Öğe Estrus synchronization during transition period, timed artificial insemination (TAI) and the effect of GnRH administration at the TAI on fertility in lactating goats(Wiley-Blackwell, 2011) Saribay, M. K.; Karaca, F.; Dogruer, G.; Ates, C.[Abstract Not Available]Öğe Preliminary report on induction of estrus with multiple eCG injections in Colored Mohair goats during the anestrus season(Elsevier Science Bv, 2009) Karaca, F.; Tasal, I.; Alan, M.This trial was conducted to evaluate the effectiveness of multiple eCG injections in the induction of estrus and pregnancy in Colored Mohair goats during the anestrus season. It was also aimed to determine total dose of eCG required for induction of estrus. Ten multiparous and lactating goats were used. The goats were randomly divided into two groups and treatments were started on May 22. Group eCG (n = 5) was treated with eCG intramuscularly for 6 days. Daily dosages of eCG from May 22 to May 27 were 300 IU, 200 IU, 200 IU, 100 IU, 100 IU and 50 IU, respectively. Goats in control group received no treatment. Blood samples were taken from animals in each of the two groups just before and after the beginning of the treatments and serum progesterone concentrations were assayed by RIA. Starting on the fourth day after the first treatments, goats were exposed to fertile bucks twice daily for 30 min to detect standing heat. The estrus goats were allowed to be mated by the bucks. Pregnancies were determined 40 days after mating by real-time ultrasonography. One goat on day 5 and three goats on day 7 exhibited behavioral estrus in eCG group (80%) after the first eCG injection. Three of them (75%) became pregnant. None of the goats in the control group exhibited behavioral estrus. Mean serum progesterone concentrations had prominent elevations indicating ovulation in eCG group, but not in control group, after 20 days from the first treatments. Progesterone concentrations of eCG group were significantly different than those of control group on days 20 and 28 (P < 0.05). The results suggest that divided multiple injections of a total 950 IU eCG are effective without progestagen pretreatment in the induction of estrus and obtaining successful pregnancy and live kids in Colored Mohair goats during the anestrus season. All rights reserved.Öğe Synchronization of estrus with short- and long-term progestagen treatments and the use of GnRH prior to short-term progestagen treatment in ewes(Elsevier, 2009) Karaca, F.; Ataman, M. B.; Coyan, K.The objective of the present study was to determine the efficacy of the synchronization of estrus using short- and long-term progestagen treatments in ewes at the onset of the breeding season, and to evaluate the effect of the exogenous GnRH administration immediately prior to short-term progestagen treatment on the reproductive performance. A total of 240 Tahirova cross-bred ewes, aged 18-24 months, and 40 rams, aged 2-4 years-old, were used in the trial. Ewes were divided equally into 3 groups (n=80 per group). Intravaginal progestagen sponges containing FGA (30 mg) were inserted in the ewes for 7 d in the FGA1 (short-term) and GnRH treatment groups, and for 12d in the FGA2 group (long-term). The ewes in the GnRH group received 10.5 mu g busereline acetate i.m. at the time of sponge insertion. Tiaprost tromethamol (PGF(2 alpha),; 0.294 mg) and eCG (400 IU) were injected i.m. on the 6th day of progestagen treatment in the GnRH and FGA1 groups, and on the 11 th day in the FGA2 group following sponge insertion. All ewes were hand-mated once at the detection of estrus. The estrous response, fertility rate, multiple birth rate and litter size recorded was 88.7, 87.3,51.6% and 1.6 in the FGA1 group, 92.5, 71.6, 50.9% and 1.5 in the FGA2 group, and 96.2, 89.6, 71.0% and 1.8 in the GnRH group, respectively. No significant difference in estrous response between the groups was recorded, but the fertility rate in the FGA1 and GnRH groups was significantly (P<0.05) higher than in the FGA2 group. The occurrence of multiple births and litter sizes were significantly (P<0.05) higher in the GnRH group, compared to both the FGA1 and FGA2 groups, with the number of single lambs being significantly (P<0.05) higher in the FGA1 (48.4%) and FGA2 (49.0%) groups than in the GnRH (29.0%) group. However, the differences recorded between any of the groups in terms of the number of twin and triplet lambs were insignificant. In conclusion, it can be said that estrous synchronization using the 12-d-FGA-eCG-PGF(2 alpha) regimen could be replaced with the 7-d-FGA-eCG-PGF(2 alpha) regime in sheep at the onset of the breeding season. However, the combination of GnRH with the latter regimen (7-d-GnRH-FGA-eCG-PGF(2 alpha)) increased the multiple birth rate and litter size in the ewes. (C) 2008 Elsevier B.V. All rights reserved.
