Öğe Association between metabolic indicators and clinical endometritis during the transition period in Brown Swiss cows(Polish Soc Veterinary Sciences Editorial Office, 2021) Yazlik, Murat Onur; Colakoglu, Hatice Esra; Pekcan, Mert; Kaya, Ufuk; Kurt, Serdal; Kuplulu, Sukru; Kacar, CihanThe objective of the current study was to evaluate the relationship between the serum macromineral, metabolite profiles, and the clinical endometritis during the transition period in Brown Swiss dairy cows. Sixty Brown Swiss dairy cows were used in the present study. Blood samples collected at d 10 (+/- 4) antepartum and 3, 10 and 30 (+/- 4) days in milk (DIM) to determine calcium (Ca), phosphorus, glucose, cholesterol, non-esterified fatty acid (NEFA) levels. Beta-hydroxybutyric acid (BHB) concentration measured during the postpartum period. Receiver operating characteristics (ROC) curves were used to determine the cow-level thresholds for the subsequent development of clinical endometritis. In addition, pairwise comparisons were made of the area under the curve (AUC) of ROC curves for the thresholds of NEFA, Ca, and glucose predicting clinical endometritis. The mean Ca concentration at 3 DIM was 8.85 +/- 0.20 mg/dL in healthy cows compared to 8.30 +/- 0.22 mg/dL in cows that subsequently developed endometritis (P < 0.05). NEFA concentrations at 10 DIM and BHB concentration at 10 and 30 +/- 4 DIM were higher (P < 0.05) in cows that subsequently developed endometritis. Serum NEFA concentration at 10 days postpartum is the best predictor for diagnosis of clinical endometritis with the AUC values of 0.741. The cows with clinical endometritis also had significantly higher values of glucose at 3 DIM and lower BCS at 10 DIM (P < 0.05). While 58.6% of the cows that developed clinical endometritis were cyclic, 64.5% were cyclic in healthy cows at 30 +/- 4 DIM. Serum NEFA concentration was the only risk factor for clinical endometritis at 10 DIM. In addition, a decrease in serum Ca at 3 DIM and increase in NEFA and BHB concentrations at 10 and 30 +/- 4 DIM may be associated with clinical endometritis and delayed resumption activity on the ovaries.Öğe Determination of zeranol residues and the serum testosterone oestrogene and progesterone levels in lambs around Ankara region(2007) Özkurt Borazan, Güzin; Karagül, Hilal; Çelik, Sefa; Ünal, Nil; Pekcan, Mert; Sel, TevhideZearalenone'un sentetik bir derivesi olan Zeranol, koyun ve sığırlarda besi performansını artırmak amacıyla anabolik madde olarak kullanılmıştır. Avrupa Birliği ülkelerinde ve Türkiye'de tüketiciye ulaşabilecek kalıntıları nedeniyle hayvanlarda kullanımı yasaklanmıştır. Yasal olmayan kullanımlarında, verilen miktar ve süre bilinmediğinden tesbiti güçtür. Bunun yanında hayvanın yası, cinsi ve bireysel farklılıklar da zeranol'ün vücuttan uzaklaşmasında etkilidir. Bu nedenle hızlı, duyarlı teknikler kullanılmalıdır ve Radioimmunoassay, referans metot olarak önerilmektedir. Ankara çevresinde çiftlik ve mezbahalardan toplanan dışkı ve doku örnelerinde zeranol analizleri ile serum örneklerinde testosteron, progesteron ve östrojen hormon analizleri Radioimmunoassay ile yapılmıştır. Toplam 153 kuzu dışkı örneği zeranol yönünden analiz edildi. Kuzularda % 22 zeranol pozitif bulundu.Öğe Effects of injectable trace element and vitamin supplementation during the gestational, peri-parturient, or early lactational periods on neutrophil functions and pregnancy rate in dairy cows(Elsevier, 2021) Yazlik, Murat Onur; Colakoglu, Hatice Esra; Pekcan, Mert; Kaya, Ufuk; Kuplulu, Sukru; Kacar, Cihan; Polat, MertThe aim of this study was to evaluate effects of injectable trace element and vitamin combination on phagocytic, oxidative burst activity of neutrophils and reproductive outcomes in dairy cows. Cows were to assigned to the following groups: (1) injectable trace element supplementation (ITES, n = 44, containing zinc, manganese, copper, selenium); (2) injectable vitamin supplementation (VIT, n = 48, containing vitamins A, D-3, E); (3) ITES + VIT = 46); and (4) control (CON, n = 44). Cows were administered four injections between 230 and 260 days of the gestational period, on day of parturition, and 30 days postpartum. Neutrophil function was assessed at 10 days before and after calving. Phagocytosis was greater in cows of the ITES + VIT group at 10 days prepartum (P < 0.05) while oxidative burst was similar among groups. There were greater non-esterified fatty acid (NEFA) concentrations in cows of the ITES+VIT group at 10 days prepartum (P < 0.05). Cows supplemented with ITES+ VIT had less SOD activity than those supplemented with ITES or vitamin during the pre- to post-partum transition period (P < 0.05). The total odds of pregnancy were greater in cows supplemented with trace element and/or vitamin (P < 0.05). In conclusion, supplementation of ITES and/or VIT resulted in an increased total pregnancy rate. Vitamin or trace element supplementation did not differ with the control group in both the prepartum and postpartum period for immune variables. There, however, was greater phagocytosis in cows supplemented with vitamin and trace elements during the prepartum period that might be related to metabolic-induced inflammation.Öğe Pathological Changes of Urinary System in a Dog with Urolithiasis and Renal Papillary Necrosis(Univ Fed Rio Grande Do Sul, 2019) Kutlu, Tuncer; Yucel-Tenekeci, Gozde; Pekcan, Mert; Kule, Emir Yengi; Kutsal, OsmanBackground: Urolithiasis is one of the important lower urinary tract diseases in dogs. Uroliths develop when urine becomes supersaturated. Struvite urolithiasis in dogs is formed when urine saturated with magnesium, ammonium and phosphate. Renal papillary necrosis is a form of nephropathy involving the necrosis of the renal papilla. The aim of this case is the evaluation of histopathological changes in kidney and urinary bladder caused by urolithiasis with sitruvite, a pathology commonly seen in dogs. Renal papillary necrosis that encountered in the case was evaluated histopathologically. We intend to discuss the relationship between urolithiasis and renal papillary necrosis. Case: The material of the case was a 7.5 year-old Rottweiler female dog that had been operated for the urolithiasis treatment and died after surgery. The animal sent to pathology department was necropsied and evaluated histopathologically. Also the uroliths removed during the surgery were sent for analysis postoperatively. Qualitative composition of uroliths was determined by a series of chemical analyses in Department of Biochemistry. X-ray diffraction was performed as well in General Directorate of Mineral Research and Exploration in the objective of result verification. Macroscopically, 30 pieces uroliths with varying size from 1 mm to 3 cm were seen in urinary bladder. They were 110 g in weight with smooth surface and round and/or ovoid shapes. After necropsy, specimens from kidney and urinary bladder were fixed in 10% neutral buffered formalin, embedded in paraffin wax, cut at 5 mu m, and stained with hematoxylin and eosin (HE). Bladder wall thickening and hemorrhage on mucosa were noticed. Kidneys had roughened surface with the capsule being difficult to peel off. Also, renal papillary necrosis, reddish in color was observed in papilla of the right kidney. Microscopically, crystal formations were observed proximal tubule lumens of kidneys. Interstitial nephritis in cortex and medulla had been noticed in both kidneys. Interstitial nephritis has been noticed in both kidneys. Hemorrhagic necrotic area demarcated by connective tissue was encountered in medulla of the right kidney. In the lamina propria of urinary bladder hemorrhage were noted. Desquamation and degeneration of lamina epithelialis and wide hemorrhage area in lamina propria were noted in urinary bladder. Results obtained from biochemical analysis were verified with x-ray diffraction method, uroliths were determined as magnesium ammonium phosphate (struvite). Discussion: Renal papillary necrosis is a form of nephropathy involving the necrosis of the renal papilla, caused by ischemia of the inner medulla of the kidney. According to the first report of renal papillary necrosis, symmetrical soft calyceal concretions and their appearances are typical lesions for diagnosis of renal papillary necrosis. The presence of crystals in the tubule lumen and the formation of struvite stone (which has the same chemical composition with calyceal concretions) established a distinct relationship between urolithiasis and renal papillary necrosis. This case showed that struvite stones can reach large volumes in urinary bladder, and caused bladder wall thickness and hemorrhages. Also, the lower urinary tract obstruction or urolithiasis should be remembered in the renal papillary necrosis together with the risk of hydronephrosis.
