    Alginate modified collagen for rapid, durable and effective biosorption of Pb (II) ions from an aqueous solution

    The aim of this study is to define the biosorption properties of Alginate, AC1 and AC2 beads composed of collagen obtained from sea urchin Diadema setosum and alginate obtained from Sargassum vulgare, for Pb (II) from aqueous solutions. Raw alginate beads (A), 2:1 alginate-modified-collagen beads (AC1), and 3:1 alginate-modified collagen beads (AC2) were successfully synthesized, characterized, and removal capacity of Pb (II) in aqueous solutions studied. The biosorption of Pb (II) ions by the synthesized beads was studied and the effect of parameters such as initial concentration, contact time, and solution pH on the biosorption of Pb (II) ions was investigated. Langmuir isotherm model provides satisfactory estimations of homogeneous monolayer biosorption capacity, the highest Pb (II) determined values being 248.76 mg/g for Alginate beads, 344.83 mg/g for AC1 beads, and 303.03 mg/g for AC2 beads. Moreover, AC1 beads were determined to be homogeneous monolayer and multilayer at two different stages. In the former stage, Pb (II) were quickly diffused to the outer surface of absorbent, and in the latter stage, ions were slowly biosorbed to the inner surface. The results obtained in this study showed that Alginate, AC1 and AC2 beads can be used effectively to remove Pb (II) ions from aqueous solutions. Another finding from this study was that the AC1 beads did not disintegration and significantly swelled in size relative to the other beads.     Bioavailability of different iron amounts and speciations and EDTA-iron complexes for Emiliania huxleyi (Prymnesiophyceae)

    The purpose of this study was to determine the effects of daily and/or initial additions of varying Fe(II) and Fe(III) amounts, in combination with or without EDTA, on Emiliania huxleyi cultures. Pertaining to this specific aim, the growth rate (mu), cell abundance (X10(4) cells L-1), total chi a concentration ([tg L-1), chi a quota (pg cell-'), chi a:C ratio, Fe content (amol cell(-1)), Fe:C and Fe:chl a (pmol:ng) ratios, as well as pH values were observed. During the experiment, iron-free seawater was obtained by using a Chelex-100 column for iron-cleaning. The results revealed that the cell densities, growth rates, chlorophyll a concentrations, Fe contents, the calculated Fe:C ratios, and pH values were higher in the cultures containing 5000 nM Fe(III) EDTA than those of 5 nM Fe(III) and Fe(II). The growth rate, cell abundance, and total chi a concentration, on the other hand, were higher with daily additions of 5 nM Fe(II) instead of 5 nM Fe(III). Furthermore, cellular Fe quota was higher in the cultures with Fe(III). Cell abundance, chlorophyll a concentration and Fe:Chl a ratio were similar in 5 nMFe(III)+ EDTA cultures (initially or daily added), possibly be due to the negative effects of EDTA on both. The growth rate, cell abundance, total chi a concentration, and Fe quota were higher in cultures with inorganic Fe(II) and Fe(IH) contents, compared to those treated with EDTA containing Fe(II) and Fe(III). E. huxleyi cells seemed not to have used EDTA-bound iron, but their own released ligand to bind iron. Unexpectedly, while cellular iron content of cultures daily supplemented with Fe(II) was low, they exhibited the highest chi a content. The cell-chl a level was higher in daily supplemented cultures, with regard to those supported only initially with iron, evidencing the importance of continuous iron supply in chi a synthesis.

    Competitive biosorption of different forms of lead [Pb(NO3)2 and Pb(CH3COO)2] on growth, biomass and proline in Spirulina platensis (Cyanophyta)

    Spirulina platensis growth parameters [chlorophyll a (chl a) and dry-wet weight] effects on proline content, lead accumulation and the combined effect of the different forms of lead [Pb (NO3)(2), Pb (CH3COO)(2)] and pH (6 to 8) were investigated for 192 h. The accumulation and form of lead were determined to be effective on growth parameters. While the highest chl a value (562.37 mu gl(-1)) and lead accumulation (58.74 mu gg(-1)) were found in medium with 30 mgl(-1) Pb (CH3COO)(2) and pH 6, the highest proline content was found in 30 mgl(-1) Pb (NO3)(2) and pH 6. This study depict an inverse relationship between lead accumulation in the test algae and low pH which suggests that proline might be produced at the expense of the material(s) required for the development of S. platensis.

    The Effect of the Environmental Factors on the Vitamin C (Ascorbic Acid), E (Alpha-tocopherol), ?-carotene Contents and the Fatty Acid Composition of Spirulina platensis

    The purpose of this study was to clarify the seasonal variation of vitamin C (ascorbic acid), E (alpha-tocopherol), beta-carotene contents and fatty acid composition of Spirulina platensis grown in the ponds in the subtropic area. While the light intensity, pH and salinity were measured daily, the temperature and dissolved oxygen were measured in day and night. The mean day temperatures were 33.9 +/- 0.4 degrees C in summer and 18.6 +/- 0.5 degrees C in winter. The mean night temperatures were measured 29.9 +/- 0.2 degrees C and 14.4 +/- 0.2 degrees C in summer and winter, respectively. The mean light intensity of 848.3 mu mol/m(2)/s was determined in summer. It was 506.26 +/- 48 mu mol/m(2)/s in winter. The vitamin C content (39.31 +/- 3.63 mg/100 g) of S. platensis grown in winter was found to be higher than in summer. The alpha-tocopherol content (6.57 +/- 1.18 mg/100 g) was higher in summer. However, -carotene contents were found to be similar both in summer and winter. Fatty acid composition was affected from the ambient factors significantly. The higher.-linolenic acid content (22.221 +/- 0.388 %) was found in summer.

    The effects of EDTA (Ethylenediaminetetraacetate) on the growth of Thalassiosira weissflogii (Bacillariophyta) in 10nM Fe(III) concentration.

    Being active in a number of biological events, iron (Fe) is essential trace metal, which is vital for a great many organisms. In an effort to find out a biological approach to the functions of Fe, which are not yet fully understood, and its available marine forms as well as to help determine the factors affecting Fe uptake, the effects of artificial ligand EDTA (Ethylenediaminetetraacetate) (100 mu M) on the growth of Thalassiosira weissflogii were studied in modified F/2 medium consisting of 10nM iron. An ambient temperature of 18 +/- 2 degrees C and lighting intensity of 80 mu mol m(-2) s(-1) were provided during the experiment. Sea water used macro nutrients and EDTA were let through Chelex-100 resin while all other items were freed from any trace metals in acid bath. Acording to the results, it was found that in T. weissflogii culture, EDTA didn't have any effect on cell number, the number of chl-a, growth rate and the pH values which increased proportional to the growth (p>0.05). It was also appeared to be a difference among cell Fe quota (p<0.05).

    This study aims to investigate the effects of two different Fe forms (Fe +2 and Fe +3) and different amounts of EDTA on the growth parameters of Spirulina platensis. During the experimental period (15 day), optical density, pH, dry weight (g L -1), chlorophyll a (?g mL -1) and phycocyanin concentration (?g mL -1) were measured on daily basis. The chlorophyll a amount of 18.30 ?g mL -1 was determined for the group containing 0.3mM Fe +3 with EDTA and 17.17 ?g mL -1 chlorophyll a content was obtained from the group containing 0.3mM Fe +2 with EDTA. Phycocyanin content was found 73.42 ?g mL -1 for the group containing 0.3mM Fe +2 with EDTA, 62.13 ?g mL -1 for control group, and 56.75 ?g mL -1 was found for the group containing 0.3mM Fe +3 with EDTA. According to these results, Fe +3 and Fe +2 forms were found to be more effective in presence of EDTA on the growth of S.platensis. © by PSP.

    The interaction effects of Cu (II), Cd (II) and Cu (II) + Cd (II) in the presence or absence of EDTA (ethylenediaminetetraacetate) on the growth of green algae Chlorella wulgaris(Chlorophyta)

    The Cu (II), Cd (II) and Cu (II) + Cd (II) ions bioaccumulation properties of Chlorella vulgaris were investigated using a batch culture as a function of in the presence/absence of EDTA and the mean growth was observed timely for 96 hours. Growth parameters of Chlorella vulgaris which are cell density, chlorophyll-a, dry-wet weight and pH values were determined for 96 hours. While the highest cell density (828.20±4.095 × 104 cell mL-1), chl a (0.42±0.017 ?g mL-1), dry weight (0.54±0.008 g L-1) and pH (10.35±0.028) were found in control group, the lowest cell density (185.89±4.509 × 10 4 cell mL-1), chl a (0.16±0.046a ?g mL -1) was in the fifth group, dry weight (0.07±0.011g L -1) in 30th and pH (5.57±0.054) in second group were found in all treatments on 24th and 48th hour of the study. Results indicated that algal growth and metal toxicity of C. vulgaris were significantly affected by the presence of EDTA, metal compounds of copper (Cu) and cadmium (Cd).
