Öğe Determination of gene escape and fruit quality characteristics in transgenic melon (Cucumis melo L. var. inodorus)(2010) Yalçın Mendi, Yeşim; Sarı, Nebahat; Akyıldız, Asiye; Solmaz, İlknur; Ünek, Ceren; Özkaya, Okan; Serçe, SedatGene escape and fruit quality characteristics of transgenic melons (Cucumis melo L. var inodorus cv. ‘Kirkagac 637’) resistant to zucchini yellow mosaic virus (ZYMV) and control plants were investigated under screenhouse conditions. No significant differences were observed between transgenic and transgenic × control genotypes, with regard to rind thickness, fruit cavity length, fruit cavity width, total soluble solids, pistil scar diameter, and peduncle length. Fruit characters, including fruit weight, fruit width, fruit length, fruit flesh thickness, and peduncle diameter were significantly different. These results indicate that transgenic × control genotypes had higher values than transgenic (T4 and T20) genotypes, regarding fruit weight, fruit width, fruit length, fruit flesh thickness, and peduncle diameter. Significant differences were not observed between transgenic (T4 and T20), control, and transgenic × control genotypes in terms of L-ascorbic acid, malic acid, citric acid, sucrose, glucose, or fructose, but differences were observed for fruit total acidity. Esters, lactones, and alcohols were aroma components, but none differed significantly between the transgenic and control genotypes. The results show that there was 100% gene escape in the control plants within 10 m of the transgenic plants, while there was 70% gene escape in plants 12.5, 15, and 17.5 m from the transgenic plants under screenhouse conditions.Öğe Rootstock potential of Turkish Lagenaria siceraria germplasm for watermelon : Plant growth, yield and quality(2012) Karaca, Fatih; Yetişir, Halit; Solmaz, İlknur; Çandır, Elif; Kurt, Şener; Sarı, Nebahat; Güler, ZehraAbstract: Th e rootstock potential of 21 bottle gourds (Lagenaria siceraria) collected from the Mediterranean region (Turkey) was investigated for watermelon with regard to plant growth, yield, and fruit quality. Th e Crimson Tide watermelon cultivar was used as a scion and 2 commercial rootstocks (L. siceraria) were also used for comparison. In greenhouse conditions, the survival rate of graft ed plants and the eff ect of rootstocks on plant growth were determined. Th e graft ed plants were planted under low tunnels in early spring and the eff ects of the rootstocks on early yield, total yield, and fruit quality were evaluated. Th e survival rates of the graft ed plants varied from 83% to 100%. Survival rates of those with commercial rootstocks and local rootstocks were similar. Plants graft ed onto 20-02, 31-09, 31-43, 35-01, and 46-03 were more vigorous than ungraft ed control plants. Compared to the control plants, graft ed plants had 37% to 80% higher plant dry weight. All of the graft ed plants except CT/Macis produced a higher yield than the control plants. Th e plants graft ed onto 6 of the local rootstocks had signifi cantly greater total yields than those graft ed onto the commercial rootstocks. Among the local bottle gourd landraces tested, the 01-16, 07-45, 20-06, 31-09, 31-15, and 46-03 were found to be promising genotypes with regard to total yield. Early yield was not signifi cantly aff ected by rootstock. Th e quality parameters of the fruits harvested from the graft ed and control plants were found to be similar except for a limited number of graft combinations. It was concluded that germplasm from Turkish bottle gourds has a high rootstock potential for watermelon with regard to the investigated parameters.Öğe Transgenik Kırkağaç 637 kavun çeşidinde morfolojik karakterizasyon(2007) Yalçın Mendi , Yeşim; Sarı, Nebahat; Solmaz, İlknur; Ünek, Ceren; Eldoğan, Selay; İpek, Muzaffer; Serçe, SedatBu çalışmada, Kabak Sarı Mozayik Virüsüne (ZYMV) dayanıklı olduğu varsayılan transgenik Kırkağaç 637 kavun çeşidinin ve kontrol bitkilerinin screenhouse koşullarında bitki özellikleri incelenmiştir. Değerlendirmeye alınan parametreler fide aşamasında kotiledon eni, kotiledon boyu ve hipokotil uzunluğu; bitki aşamasında ise bitki boyu, ana gövde çapı, boğum sayısı, yaprak sapı uzunluğu, yaprak genişliği ve yaprak boyudur. Yapılan analizler sonucunda transgenik bitkilerde kotiledon eni ve hipokotil uzunluğunun, transgenik olmayan bitkilere göre daha kısa olduğu bulunmuş, kotiledon boyları bakımından ise istatistiksel olarak bir fark saptanmamıştır. Transgenik ve transgenik olmayan bitkiler arasındaki bitki boyu (%1 önem seviyesinde), ana gövde çapı, boğum sayısı ve yaprak sapı uzunluğu da (%5 önem seviyesinde) istatistiksel olarak farklı bulunmuştur.
