Gökkuşağı alabalığı (Oncorhynchus mykiss) larvalarının proteaz aktiviteleri üzerine farklı yem hammaddelerinin inhibisyon etkilerinin belirlenmesi

Özet
Bu çalışmada, hayvansal yem hammadde kaynaklarından balık unu, balık protein hidrolizatı ile bitkisel yem hammadde kaynaklarından soya unu, ayçiçek tohumu küspesinin ve buğday glüteninin gökkuşağı alabalığı (Oncorhynchus mykiss Walbaum, 1792) larvalarının proteaz aktiviteleri üzerine inhibisyon etkileri araştırıldı. Çalışmada, gökkuşağı alabalığı larvalarında en düşük proteaz aktivitesi, yumurtadan çıkışı takiben 5. günde 13,93±2,45 U/mg protein olarak belirlenirken, en yüksek proteaz aktivitesi ise 307.83±2.14 U/mg protein olarak yumurtadan çıkışı takiben 35. günde belirlendi. Proteaz aktivite değerlerinde 5. günden 35. güne kadar artış tespit edilirken, 35. günden 50. güne kadar ise bu değerlerin azaldığı belirlendi. Proteaz aktivite değerleri arasındaki farklılıkların istatistiksel olarak önemli olduğu tespit edildi (p<0.05). Hayvansal yem hammadde kaynaklarından balık unu için en düşük ve en yüksek proteaz inhibisyon yüzdeleri sırasıyla % 13,93±2,45 (5. gün) ve % 67.33±3.82 (15. gün) olarak belirlenirken, balık protein hidrolizatı için ise proteaz inhibisyon yüzdeleri en düşük % 18.69±6.95 olarak 40.gün, en yüksek % 61.53±6.29 olarak 10. gün tespit edildi. Proteaz inhibisyon yüzdeleri, bitkisel yem hammadde kaynaklarından soya unu için en düşük % 51.36±14.79 (5. gün) ve en yüksek % 91.49±2.93 (15. gün) olarak, ayçiçeği tohumu küspesi için en düşük % 24.37±3.93 (35. gün) ve en yüksek % 85.68±2.36 (15.gün) olarak, buğday gluteni için en düşük % 45.62±1.19 (50. gün) ve en yüksek % 88.36±2.49 (15. gün) olarak belirlendi.
In this research, inhibition effect of different animal (fish meal and fish protein hydrolisate) and plant (soybean meal, sunflower seed pulp and wheat gluten) raw material sources on protease activity of rainbow trout (Oncorhynchus mykiss) larvaes were investigated. In rainbow trout larvaes, while the lowest protease activity (11.45±0.99 U/mg protein) was determined at 5th day, the highest protease activity (307.83±2.14 U/mg protein) was determined at 35thday following hatching. Protease activity value showed an increase between 5th and 35th days but an decrease was determined at this value following 35th day until 50th day. Statistically significant differences were determined among the protease activity values (p<0.05). The lowest and highest protease inhibition percentages were determined respectively as 14.93±2.45 % (5th day) and 67.33±3.82 % (15th day) for the fish meal while the same percentage values were determined respectively as 18.69±6.95 % (40th day) and 61.53±6.29 % (10th day) for the fish protein hydrolizate. The lowest and highest protease inhibition percentages were determined respectively as 51.36±14.79 % (5th day) and 91.49±2.93 % (15th day) for the soybean meal, 24.37±3.93 % (35th day) and 85.68±2.36 (15th day) for the sunflower seed pulp and 45.62±1.19 % (50th day) and 88.36±2.49 % (15th day) for the wheat gluten.
