Streptozotosin ile diyabet oluşturulan sıçanlarda zeytin yaprağı ekstresinin glikoz metabolizmasında etkili olan karaciğer enzimleri üzerine etkisinin incelenmesi

Özet
Streptozotosin ile Diyabet Oluşturulan Sıçanlarda Zeytin Yaprağı Ekstresinin Glikoz Metabolizmasında Etkili Olan Karaciğer Enzimleri Üzerine Etkisinin İncelenmesi Çalışmamızda streptozotosin (STZ) ile diyabet oluşturulan sıçanlarda zeytin yaprağı ekstresinin (OLE) glikoz metabolizmasında etkili olan karaciğer enzimleri üzerine etkisinin incelenmesi amaçlandı. 32 adet WistarAlbino cinsi sıçan 4 gruba ayrıldı. Grup 1 (Kontrol), Grup 2 (STZ ile diyabet oluşturulan grup), Grup 3 (STZ+Zeytin yaprağı ekstresi uygulanan grup) ve Grup 4 (OLE+STZ uygulanan grup). Kullandığımız zeytin yaprakları Akdeniz bölgesinin Hatay ilinden toplandı. 3. gruptaki hayvanlara 6 hafta boyunca 0.5g/kg zeytin yaprağı ekstresi oral gavaj yöntemi ile verildi. 4. gruba STZ uygulamasından 2 gün öncesinden başlanarak 6 hafta boyunca her gün 0.5g/kg OLE verildi. Cerrahi uygulama öncesi kan glukoz değerleri ölçüldü ve glikoz metabolizmasında etkili olan hekzokinaz (HK), piruvat kinaz (PK), glukoz 6 fosfataz (G6P), fruktoz-1,6-bifosfataz (FBP1), glukoz 6 fosfat dehidrogenaz (G6PD) enzimlerinin uygun kitlerle ölçümleri yapıldı. Bu çalışmamızdan elde edilen bulgular, STZ ile oluşturulan deneysel diyabet modelinde glukoz metabolizması ile ilgili enzimlerin miktarında bozulmalar meydana getirdiğini göstermiştir. OLE uygulanmasının enzim düzeylerini kontrol grubu değerlerine döndürmeye yardımcı olarak diyabet tablosunda iyileşme sağladığı söylenebilir. Zeytin yaprağı ekstresinin önceden alınmasının da olumlu sonuçlar sağladığını düşünmekteyiz.
Investigation of The Effects of Olive Leaf Extract on Liver Enzymes In Streptozotocin-Induced Diabetic Rats The aim of this study was investigating effects of olive leaf extract (OLE) on liver enzymes which are effective in glucose metabolism in streptozotocin-induced diabetic rats. 32 male Wistar albino rats were classified 4 groups as group1: control (C); group2: streptozotocin-induced diabetic group (STZ); group3: STZ+OLE; group4: OLE+STZ group. We used olive leaf which were collected from Hatay city. 0.5 g/kg/day of OLE was given orally to STZ+OLE group for 6 weeks. 0.5 g/kg/day of OLE which was started to give before 2 days from streptozotocin-induced diabetes, was given orally to OLE+STZ group every day for six weeks. At the end of study, glucose levels of rats were measured afterwards rats were sacrificed and blood samples were collected for analyses of hexokinase (HK), pyruvate kinase (PK), glucose-6-phosphatase (G6P), fructose-1,6- bisphosphatase (FBP1), glucose-6-phosphate dehydrogenase (G6PD) which are effective in glucose metabolism. These results of study were showed that enzymes levels associated with glucose metabolism were deteriorated in STZ-induced diabetic model. It was suggested that treatment of OLE may recovery diabetes by helping to return enzyme levels to control group. We think pretreatment of OLE may give beneficial results.
