Hatay yöresindeki ochthebius (Hydraenidae, Coleoptera) türlerinin nükleer ve mitokondriyal rRNA'lar hedeflenerek ortaya çıkarılması

Özet
Bu çalışmada morfolojik özelliklerine bakılarak tanımlanan Hatay yöresinde ve Doğu Anadolu'da yaşayan 17 su böceği türünün ( Ochthebius) Cytokrom C Oksidaz Alt Ünite 1 (COX) genini DNA dizilimleri moleküler çalışmalar sonucu elde edilmiştir. İncelenen sucul böcekler, farklı bitki örtüsüne sahip ve farklı yüksekliklerde bulunan göl, bataklık , su birikintileri, gölet, vb. habitatlardan 2000 - 2013 yılları arasında yapılan arazi çalışmaları ile toplanmıştır. Örnekler tür teşhisi yapıldıktan sonra -20 °C'de soğutucuda petri kapları içerisinde saklanmıştır. Daha sonra COX 1 bölgesi evrensel primer kullanılarak PCR yoluyla çoğaltılmıştır. Çoğaltılan DNA bölgelerinin baz dizin analizleri yapılmıştır. Bu filogenetik analizler 3 farklı algoritma; maximum parsimony , maximum likelihood, Neighbor-joining programıyla değerlendirilmiştir. Değerlendirme sonucu Hydraenidlere ait alt türlerin parsimonik ağacı oluşturulmuştur. Bu çalışmada Hydraenidler üzerinde denenerek elde edilen verilerle, morfolojik ve anatomik çalışmalara dayanan tekniklerle daha doğru sonuçlara götüren moleküler sistematik yöntemlerle kıyaslanarak tür teşhisi için yeterli olup olmadığı gözlenmiştir.
With this study, DNA sequences of Cytochrome C Oxydase Subunit I (COX I) gene were worked out bye molecular proceedings for morphologically identified seventeen Ochthebius species (Hydraenidae, Coleoptera) in Hatay province and eastern Anatolia region. These species were collected during field work carried out in 2000 and 2013 at localities with various altitudes and vegatation in Hatay and eastern Anatolia region. Samples collected were first identified species to their morphological features prious to be placed on plates in the freezer at the -20 0C. Afterwords, in order to determine their COX I section, DNA isolation was rendered with appropriate methods. PCR method was employed to increase the COX I section of the samples with the help of universal primers. Then basic sequence analysies were made for the increased with 3 different algorithms, namely maximum parsimony, maximum likelihood and Neighbor-joining programmes. Based on this assesment the parsimonic tree was to reveal the resemblance logic between species pertaining in Ochthebius species. In the light of the findings from this study, it was established that phylogenetic analysies for these seventeen Ochthebius species, which was not studied with moleculer data in our country, provided universally valid and more reliable information when compared with morphological data in identifying species.
