Özet
Bu tez çalışmasında, kuraklık ve kuraklık-NO (nitrik oksit) etkileşimlerinin buğday fidelerindeki (Triticum aestivum L. cv. Sultan, Yüreğir, İkizce ) etkileri incelenmiştir. Sürgün boyu, sürgün taze ve kuru ağırlığı, klorofil a (kl a), klorofil b (kl b), toplam klorofil (kl a+b) miktarı, prolin ve malondialdehit (MDA) birikimi, katalaz (CAT) ,glutatyon redüktaz (GR) ve askorbat peroksidaz (APX) antioksidan enzim aktiviteleri analiz edilmiştir. Kuraklıkta aktivitesi artan genlerin No Apical Meristem (NAM) ATAF1/2 CUC2 (NAC) ve Triticum aestivum Lipid Transfer Proteini (TaLTP1) ifadesi gerçek zamanlı polimeraz zincir reaksiyonu (RT-PCR) tekniği ile belirlenmiştir. Sonuç olarak, kuraklık uygulaması sonucu sürgün boyu ve taze ağırlığının her üç çeşitte de azaldığı gözlemlenmiştir. Sürgün kuru ağırlığının Sultan ve İkizce türlerinde azalırken Yüreğir türünde arttığı gözlenmiştir. Kuraklık uygulamasının sürgün fidelerinin belirli pigment miktarlarında (kl a, kl b ve toplam kl) azalmaya neden olurken, kuraklık-NO uygulamasının her üç parametrede, bir miktar iyileşmeye yardımcı olduğu görülmüştür.Sultan fidelerinde antioksidan enzim aktivitelerinin (CAT, GR ve APX) stresine bağlı olarak arttığı, Yüreğir ve İkizce fidelerinde kuraklıkta CAT aktivitesi azalırken, GR ve APX aktivitesi arttığı tespit edilmiştir.Yaptığımız çalışmada gen verileri incelendiğinde strese bağlı olarak kontrole göre NAC ve TaLTP1 gen ifadelerinin her üç çeşitte de arttığı, bu artışın kuraklığa duyarlı Sultan çeşidinde daha fazla olduğu saptanmıştır.
In this thesis, effect of drought and drought-NO (nitric oxide) interactions on three wheat seedlings (Triticum aestivum L. cv. Sultan, Yüreğir, İkizce) was studied. Length of shoot, wet and dry weight of shoot, chlorophyll a (chla) and b (chlb), total chlorophyll (chl a+b) amount, proline and malondialdehyde (MDA) accumulation, also antioxidant enzyme activities (catalase (CAT), glutation reductase (GR) and ascorbate peroxidase (APX) were analysed and as well as gene expressions which occurred in drought stress were determined by real-time polymerase chain reaction (RT-PCR) technique. As a result, in all three types of seedlings, decrease in both length of shoot and wet weight of shoot were observed. Also, whereas in Sultan and İkizce types decrease in dry weight of shoot was observed, in Yüreğir types increase is observed.When effect of drought types were analysed in detail, it was noticed that drought stress application trigger decrease in specific pigment amounts of shoots seeds (chl a, chl b and chl a+b), while drought-NO triggered increase In all three types of seedlings, increase in antioxidant enzyme activities (CAT, GR and APX) associated with drought stress was detected. Moreover, increase of MDA and free proline accumulation in wheat seedlings that were exposed to drought was detected.Furthermore, in all three types of seedlings, increase of expressions of NAC and TaLTP1 genes was observed, even much more level in the drought sensitive variety Sultan.
