Özet
Bu araştırmada, buğday (Triticum aestivum L. cv. Dağdaş, Doğankent) fidelerinde su baskını, kuraklık ve tuz streslerinin etkileşimleri incelenmiştir. Sürgün boyu ve sürgün kuru ağırlığı, klorofil a (kl a), klorofil b (kl b), toplam klorofil (kl a+b), katalaz (CAT) askorbat peroksidaz (APX) ve glutatyon redüktaz (GR) antioksidant enzim aktiviteleri, prolin miktarı, malondealdehit ve bu abiyotik streslerin etkileşimlerinde çalışan CHL, TaSRHP ve TaZnFP genlerinin ifadesi belirlenmiştir.Araştırmamızda kullandığımız Dağdaş ve Doğankent buğday çeşitlerinde kontrol grubuna kıyasla stres uygulanan gruplarda stres faktörlerinin sürgün gelişimini olumsuz yönde etkilediği gözlenmiştir. Abiyotik streslerin etkisiyle klorofil a, klorofil b ve toplam klorofil miktarlarında da bir azalma meydana gelmiştir. Katalaz, glutatyon redüktaz ve Askorbat peroksidaz aktivitelerinde ise strese bağlı olarak kontrole göre artışlar meydana gelmiştir. Gen verilerimiz incelendiğinde ise stres faktörlerine bağlı olarak kontrole göre TaSRHP geninde artış fakat CHL geninde azalma meydana geldiği görülmüştür. TaZnFP proteininde ise hiçbir aktivite gözlenmemiştir
In the present study, drought, water and salt stres their interactions were investigated on wheat seedlings (Triticum aestivum L. cv. Dagdas and Doğankent). Shoot length and shoot dry weight, chlorphyl a and b, total chlorophyl (a+b) amount, catalase, glutation reductase enzyme activity as well as gene expressions during abiotic stres were investigated.Stress negatively affected shoot develeopment on wheat cultivars Dagdas and Doğankent used in this study. In addition, amount of chlorophyll a, chlorophyll b and total chlorophyll also decreased with abiotic stress appilications. Catalaz (CAT), glutation reductase (GR) and APX activities were higher than the control treatment. All of this stress treatments increased the enyzme activities. When analyzed genes stress factors depending on our data TaSRHP gene increased compared to control occurred but it was observed that a decrease in gene CHL. TaZnFP protein was observed no activity
