Forklorfenuron (FCF) sitokininler grubuna dahil olan ve dünya çapında tarımsal uygulamalarda yaygın olarak kullanılan bir bitki gelişim düzenleyicisidir. Yaygın kullanım alanına sahip olmakla birlikte, FCF'nin genotoksik potansiyeliyle ilgili bilgiler oldukça kısıtlıdır. Bu nedenle, bu çalışmada FCF'nin insan lenfositleri üzerindeki potansiyel genotoksik etkisinin in vitro kromozom anormalliği (KA) testi ile araştırılması amaçlanmıştır. Kültürler, 0.25, 0.50, 1.00 ve 2.00 μg/ml konsantrasyonlarındaki FCF ile 24 ve 48 saat süre ile muamele edilmiştir. Çalışmada her deneye paralel olarak negatif kontrol ve pozitif kontrol kullanılmıştır. FCF, yüksek iki konsantrasyonda (1.00 ve 2.00 μg/ml) ve her iki uygulama periyodunda (24 ve 48 saat) KA oluşumunu uyararak anormal hücre yüzdesini negatif kontrole göre önemli derecede arttırmıştır. Ayrıca, toplam KA/hücre oranı, 0.50, 1.00 ve 2.00 μg/ml konsantrasyonlarındaki FCF uygulanan tüm kültürlerde negatif kontrole göre önemli derecede yüksek bulunmuştur. Fakat bu artışlar hiçbir uygulamada pozitif kontrol kadar olmamıştır. Ayrıca, FCF'nin tüm konsantrasyonlarda ve uygulama sürelerinde (24 saatlik uygulamada en düşük konsantrasyon hariç) mitotik indeksi negatif kontrole göre önemli derecede düşürerek sitotoksik aktiviteye sahip olduğu bulunmuştur. Sonuçlar, FCF'nin insan lenfositleri üzerinde genotoksik ve sitotoksik potansiyele sahip olabileceğini göstermektedir.
Forchlorfenuron (FCF) is a plant growth regulator that belongs to the group of cytokinin and is widely used in agricultural applications around the world. Despite its widespread use, information on the genotoxic potential of FCF is very limited. Therefore, in this study, it was aimed to investigate the potential genotoxic effect of FCF on human lymphocytes by in vitro chromosomal aberration (CA) test. Cultures were treated with FCF at concentrations of 0.25, 0.50, 1.00 and 2.00 μg/ml for 24 and 48 hours. In the study, negative control and positive control were used in parallel with each experiment. FCF, at two high concentrations (1.00 and 2.00 μg/ml) and in both treatment periods (24 and 48 hours), significantly increased the percentage of abnormal cells compared to the negative control by stimulating CA formation. In addition, the total CA/cell ratio was significantly higher in all cultures treated with FCF at 0.50, 1.00 and 2.00 μg/ml concentrations compared to the negative control. However, these increases were not as high as the positive control in any application. In addition, FCF was found to have cytotoxic activity at all concentrations and treatment times (except for the lowest concentration at 24-hour), significantly reducing the mitotic index compared to the negative control. The results indicate that FCF may have genotoxic and cytotoxic potential on human lymphocytes.
