Gebelikle ilişkili glikoprotein test kitinin keçilerde erken gebelik tanısındaki etkinliğinin belirlenmesi

dc.description.abstractSunulan çalışmada, gebelikle ilişkili glikoproteinlerin (PAGs) kan serumundan tespitine yönelik hazırlanan ticari hızlı görsel gebelik test kitinin keçilerde erken gebelik tanısındaki etkinliğinin ve güvenilirliğinin araştırılması amaçlandı. Çalışmada sağlıklı ve fertil, 3 ila 5 yaşlı 150 baş Halep ırkı keçi kullanıldı. Keçilere üreme mevsimine geçiş döneminde östrus senkronizasyon uygulaması yapıldı. Östrüsteki keçilere doğal aşım yaptırıldı. Aşımlarından sonraki 28. günde keçilerden kan örnekleri alındı. Gebelik tanısı için aşımlardan 50 gün sonra keçilere transabdominal ultrasonografi uygulandı. Alınan kan örneklerinden progestron ölçümü (Direct Chemiluminescence yöntemi; ADVIA Centaur® ReadyPackTM, Siemens, USA) ve hızlı görsel gebelik testi (IDEXX Rapid Visual Pregnancy Test Kiti; Westbrook, Maine, USA) yapıldı. Ultrasonografik muayene sonuçları referans değer olarak kullanıldı. Serum progesteron ölçümü ve görsel PAG-ELISA kitinin sensitivite (duyarlılık), spesifite (özgüllük), pozitif prediktif değer (pozitif tahmin değer), negatif prediktif değer (negatif tahmin değer) oranlarının sırasıyla %91.18, %93.90, %92.54, %92.77 ve %94.12, %80.49, %80, %94.29 olduğu belirlendi. Çalışmada progesteron konsantrasyon bulgularının ultrasonografi bulguları ile uyumunun çok iyi (kappa=0.852), görsel PAG-ELISA bulgularının ultrasonografi bulguları ile uyumunun iyi (kappa=0.735) olduğu tespit edildi. Ayrıca progesteron konsantrasyon bulgularının görsel PAG-ELISA bulguları ile uyumu da iyi (kappa=0.748) olarak belirlendi. Serum progesteron ölçümü ve görsel PAG-ELISA kitinin doğru tanı koyabilme yetenekleri de sırasıyla %92.67 ve %86.67 olarak bulundu. Sonuç olarak; keçilerde görsel PAG-ELISA testinin de progesteron ölçüm sonuçlarına yakın oranda doğru sonuç verdiği ve pratikte görsel PAG-ELISA ile erken gebelik teşhis yönteminin tek başına veya diğer yöntemlerle kombinasyon halinde kullanılabileceği kanısına varıldı.en_US
dc.description.abstractIn this study it was aimed to investigate the efficacy and reliability of the commercial rapid visual pregnancy test kit prepared for the detection of pregnancy-related glycoproteins (PAGs) in blood serum for the diagnosis of early pregnancy in goats. For this purpose, healthy and fertile, 3-5 years old 150 Damascus goats were used. For induction of Oestrus, synchronization was applied to the goats during transition period to the breeding season and natural mating was performed to the goats in the oestrus. After mating, blood samples were taken from all goats on day 28 and transabdominal ultrasonography was done for diagnosis of pregnancy on day 50. With the blood samples, progesterone measurements (Direct Chemiluminescence method; ADVIA Centaur® ReadyPackTM, Siemens, USA) and visual pregnancy test (IDEXX Rapid Visual Pregnancy Test Kit; Westbrook, Maine, USA) were performed. For serum progesterone measurement and visual PAG-ELISA kit, the sensitivity, specificity, positive predictive value, negative predictive value rates were determined as 91.18%, 93.90%, 92.54%, 92.77%, and 94.12%, 80.49%, 80%, 94.29%, respectively. In the study, the agreement between concentration of progesterone and ultrasonography was strong (kappa=0.852), the agreement between PAG-ELISA and ultrasonography was moderate (kappa=0.735). Additionally, the agreement between concentration of progesterone and PAG-ELISA was moderate (kappa=0.748). Furthermore, the accuracy of serum progesterone measure and visual PAG-ELISA kit were 92.67% and 86.67%, respectively. As a result, it was concluded that the visual PAG-ELISA test in goats gives an accurate result close to the progesterone measurement results and in practice, the visual PAG-ELISA method for early pregnancy detection can be used alone or combined with other methods.en_US
