Dipironun antidepresan-benzeri etkisinin zorunlu yüzdürme testiyle araştırılması

Özet
Analjezik-antipiretik ilaç olarak kullanılan dipironun özellikle COX-3 inhibisyonu olmak üzere santral prostaglandin sistemi, opiaterjik ve kannabinoid sistemler, sitokin düzeyleri, TRP kanal- ları ve inici serotonerjik-noradrenerjik yolaklar gibi çeşitli meka- nizmalar üzerinden etkilerini gösterdiği ortaya konmuştur. Duygu durum bozukluklarında da tüm bu mekanizmaların fonksiyonu olduğu belirlenmiştir. Ayrıca ağrı ve depresyonun çoğu olguda birlikte görüldüğü bilinmektedir. Bu bilgiler ışığında çalışmamız- da dipironun antidepresan benzeri etkisi ve depresyonda kulla- nılan ilaçların etkisini artırıcı özelliği olup olmadığını araştırmayı amaçladık. Deneylerde depresyon benzeri etki, erkek fareler kul- lanılarak zorunlu yüzdürme testinde incelendi. Silindir şeklindeki, cam kapta altı dk. yüzdürülen hayvanların suda hareketsiz kal- ma süreleri ölçüldü. Bu süre depresyon-benzeri etkinin göster- gesi olarak kabul edildi. Kullanılan ilaçlarla bu sürenin azalması antidepresan-benzeri etki olarak yorumlandı. Deneylerde dipiron 150, 300 ve 450 mg/kg ip ve Floksetin 20 mg/kg dozunda oral gavaj yoluyla uygulandı. Çalışmamızda, dipironun farklı üç dozu- nun uygulandığı farelerin suda hareketsiz kalma süreleri, kontrol grubuyla karşılaştırıldığında anlamlı olarak azaldığı görüldü. Di- pironun 150 mg/kg dozundaki etkisinin, Floksetin 20 mg/kg do- zuyla aynı olduğu belirlendi. Her iki ilacın birlikte kullanılmasıyla oluşan etki, tek başına kullanıldıklarında elde edilen etkiden farklı değildi. Bu çalışmada, dipironun zorunlu yüzme testinde flokse- tin gibi antidepresan-benzeri etki gösterdiği belirlendi. İki ilacın birlikte kullanılmaları sonucu floksetinin etkisinin değişmediği görüldü. Bütün bu bulgular, dipironun antidepresan-benzeri etki- sinin olduğunu telkin etmektedir. Dipironun antidepresan-benzeri etkisinin diğer testlerle ve ilaçlarla karşılaştırılarak araştırılması ve etki mekanizmasının aydınlatılması gerekmektedir. Bu bulgu- ların, gelecekte depresyon ve ağrının birlikte seyrettiği olguların tedavilerine yeni katkılar sağlayacağı düşünülmektedir.
Dipyrone is an analgesic-antipyretic drug with especially COX-3 inhibition on central prostaglandin system, proven effects on opiatergic and cannabinoid systems, cytokine levels, TRP chan- nels and descending serotonergic-noradrenergic pathways. The functional roles of all these mechanisms have been established in mood disorders. It is known that pain and depression are frequ- ently seen together. In the light of these information in our study we aimed to investigate whether dipyrone has an antidepressant- like effect and an antidepressant-medication-enhancing feature. Forced swimming test was utilized to measure depression-like effects of dipyrone using male mice. Immobility time of the ani- mals was considered indicative of the depression-like effect on animals which were floated for six minutes in cylinder-shaped glass containers. The reduction in the duration of immobility with the drugs used was interpreted as an antidepressant-like effect. In the experiments dipyrone was administered via intraperitone- al route at doses of 150, 300 and 450 mg/kg and fluoxetine 20 mg/kg by oral gavage. It was observed that the immobility time of the mice in which all doses of dipyrone were administered dec- reased significantly when compared with the control group. The effect of 150 mg/kg doses of dipyrone was determined to be the same with fluoxetine. When the two drugs were used together, the effects of the drugs did not differ from using the drugs indi- vidually. In this study, it was determined that dipyrone showed antidepressant-like effects comparable to fluoxetine, and when both drugs were used together the effect of fluoxetine in forced swimming test did not change. All these findings suggest that dipyrone has an antidepressant-like effect. Antidepressant like effects of dipyrone in comparison with other drugs should be in- vestigated with further testing. It is thought that this finding will contribute further to the treatment protocols of cases coursing with depression and pain.
