Özet
Bu çalışmada, subklinik mastitisli inek sütlerinden izole edilen Staphylococcus aureus izolatlarının toksin genleri ve koagülaz gen polimorfizmi yönünden karekterizasyonunun yapılması amaçlandı. İncelenen 130 S. aureus izolatının 60 (% 46.2)'ı bir veya birden fazla toksin geni yönünden pozitif bulundu. Pozitif bulunan izolatların 18'inde seg - sei ve sec, 14'ünde sed - sej, 6'sında tst, 2'sinde sed - sej - seg - sei, 1'er izolatta ise sec - seg - sei ve sec - tst genleri tespit edildi. sea, seb, see ve seh genleri yönünden ise tüm izolatlar negatif bulundu. S. aureus izolatlarının coa genine dayalı PZR analizinde 127 izolatta moleküler büyüklükleri 730 - 970 bp arasında değişen tek band ve 3'ünde ise moleküler büyüklüğü 810 + 1050 bp olan iki band tespit edildi. coa-PZR ürünlerine göre izolatlar 5 farklı tip (A, B, C, D, E) oluşturdu. coa pozitif PZR ürünlerinin AluI restriksiyon enzimi ile kesilmesi sonucu toplam 9 farklı restriksiyon profili elde edildi. Tip B (B1 ve B2) ve Tip C (C1 ve C2)'ye ait izolatlar iki, Tip D'ye ait izolatlar ise 3 alt tipe (D1, D2 ve D3) ayrıldı. Tip B (55 izolat) ve Tip E (42 izolat) en dominant tipler olarak belirlendi. Bu çalışmada, yaygın genotiplere ait izolatların büyük kısmının toksin genleri yönünden pozitif olduğu tespit edilmesine rağmen, C, D1 ve D2 genotipine ait izolatlarda toksin genlerinin varlığı belirlenemedi. Sonuç olarak, (i) Hatay ilinde subklinik inek mastitislerinin farklı S. aureus genotipleri tarafından meydana getirildiği, ancak bir veya iki genotipin dominant olduğu, (ii) dominant genotiplerin enterotoksijenik olduğu, bu genotiplerin bir veya birden fazla enterotoksin geni taşıdığı, (iii) S. aureus'un sığırlarda neden olduğu meme içi infeksiyonların patogenezinin anlaşılması için farklı bölgelerden toplanan çok sayıda izolat ile yapılacak daha kapsamlı çalışmaların yapılması gerektiği sonucuna varılmıştır.
The aim of this study was to characterize Staphylococcus aureus isolates obtained from subclinic bovine mastitic milk samples by coagulase gene polymorphism and toxin genes. Out of 130 S. aureus isolates, 60 isolates (46.2 %) were found positive for having one or more toxin gene. Among positive isolates, it has been determined that 18 of the isolates containing seg - sei, 18 of the isolates containing sec, 14 of the isolates containing sed - sej, 6 of the isolates containing tst, 2 of the isolates containing sed - sej - seg - sei, 1 of the isolates containing sec - seg - sei, and 1 of the isolates containing sec - tst genes. No sea, seb, see and seh gene was detected among the isolates tested. PRC analysis of S. aureus isolates based on coa gene, a single band with a molecular weight of 730-970 bp was observed form 127 isolates, and two bands were observed in 3 isolates with molecular weight ranging from 810 + 1050 bp. Isolates produced 5 different type (A, B, C, D, E) according to coa-PRC products. Nine different restriction profile were obtained when coa positive PRC products were being cut with AluI restriction enzymes. While Type B and Type C isolates were classified into two subgrups, as B1, B2, C1, C2; Type D isolates classified into three subgrups as D1, D2, D3, respectively. Type B (55 isolates) and Type E (42 isolates) were determined as the most dominant types. In this research, although it was determined that most of the isolates obtained from common genotypes were toxin gene positive, no toxin gene was determined from isolates obtained from C, D1, and D2 genotypes. It was concluded that (i) subclinic cow mastitis caused by different S. aureus genotypes in Hatay and only one or two of the genotypes were dominant, (ii) dominant genotypes were enterotoxigenic and have one or more that one enterotoxin genes (iii) it is suggested that more comprehensive studies with more isolates obtained from different regions to determine patogenicity of bovine mastitis caused by S. aureus need to be performed.
