Özet
Tez kapsamında ekstrem halofilik arke suşları ile potansiyel krom, gümüş, çinko ve nikel nanopartiküllerinin sentezlenmesi amaçlanmıştır. Standart suşlar ile tuzlu toprak ve su örneklerinden izole edilmiş toplam 113 haloarkeal suş %20 NaCL ve farklı metal tuzları (0,5 ve 1 mM) içeren kompleks katı ve sıvı ortamlarda üreme yetenekleri yönünden taranarak potansiyel nanopartikül üretici aday izolatların seçimi yapılmıştır. Seçilen izolatların sıvı ortamda 1,5 mM'a kadar artan konsantrasyonlardaki metal tuzlarına adaptasyonu sağlanarak metalik nanopartikül üretimi yapılmıştır. Çalkalamalı kültür ortamında hücre içi üretilen nanopartiküllerin karakterizasyonu XRD ve SEM-EDS ile belirlenmiştir. Biyosentezi yapılan gümüş nanopartiküllerinin yüzey merkezli kristal yapılara sahip olduğu ve SEM-EDS analizleri sonucunda herbir metalin elementel formunun mevcut olduğu ve üretilen nanopartiküllerin çekim gücüne bağlı olarak homojen dağılım gösterdiği bulunmuştur. Bu çalışma ile ilk olarak gümüş haricinde krom, nikel ve çinko metalik nanopartiküllerinin fiziksel ve kimyasal yöntemler haricinde çevre dostu biyojenik sistemlerden ekstrem halofilik arkelerle üretilebileceği gösterilmiştir.
It is aimed to synthesize potential chromium, silver, zinc and nickel nanoparticles with halophilic archaea, within the scope of the thesis. The potential nanoparticle producer candidate isolates were selected by screening a total of 113 haloarcheal strains isolated from saline soil and brine samples and standart strains for their ability to grow in complex agar and broth media containing 20% NaCL and different metal salts (0.5 and 1 mM). Metallic nanoparticles were produced by adapting the selected isolates to metal salts at increasing concentrations up to 1.5 mM in broth medium. The characterization of nanoparticles produced intracellularly under agitated conditions was determined by XRD and SEM-EDS. XRD analyzes of biosynthesized silver nanoparticles showed that they have face-centered crystal structures. SEM-EDS analaysis revealed the formation of well-dispersed nanoparticles and the presence of elemental form of each metal was confirmed. To the best of our knowledge, this is the first report on the synthesis of chromium, nickel and zinc nanoparticles by haloarchaea from environmentally friendly biogenic systems, apart from physical and chemical methods.
