Özet
Amaç: Bu çalışmada akustik travmaya bağlı ani işitme kaybında Platelet Zengin Plazma (PRP) ile geleneksel tedavilerin işitmenin iyileşmesine olan katkılarını deneysel düzeyde test edilmesi amaçlandı. Gereç ve Yöntem: Çalışmada 30 adet sağlıklı erişkin rat kullanıldı. Çalışma öncesi deney hayvanlarının otoakustik emisyon (OAE) ölçümleri yapıldı. OAE ölçümleri yapılan tüm ratlar 60 dakika süre ile, 4 kHz frekansında 95 dB saf ton sese maruz bırakıldı. Deney hayvanlarının 24. saatte OAE'leri tekrar ölçülerek akustik travma oluştuğu tespit edildi.10'lu şekilde 3 gruba ayrılan ratlardan ilk grup kontrol grubu, 2. grup 2 gün arayla 5 doz yaklaşık 0,5 ml intratimpanik steroid enjekte edilen ve 3. grup yine 2 gün arayla yaklaşık 0,5 ml intratimpanik PRP enjekte edilen grup olarak belirlendi. Yaklaşık 10 gün sonra yapılan OAE ölçümleri değerlendirilerek verilen tedavilerin, tedavi verilmeyen gruba göre karşılaştırılması ve istatiksel olarak anlamlı fark olup olmadığı değerlendirildi. Bulgular: DPOAE ölçümlerinin gün gün, frekans frekans ölçümlerinden elde edilen bulgular istatistiksel olarak karşılaştırılarak anlamlı sonuç alınıp alınmadığı kontrol edildi. Hiç tedavi almayan akustik travmalı ratlara göre PRP ve prednol ile tedavi alanlarda daha iyi sonuçların elde edildiği, PRP ile prednol grupları arasında ise yakın tedavi etkinlik sonuçları tespit edildi. Sonuç: Bu üç grubun dört frekansta, çeşitli günlerdeki DPOAE ölçümlerinin karşılaştırmasında bazı gün ve frekanslarda anlamlı istatistiksel fark mevcutken bazılarında anlamlı fark alınamamıştır. Bunun nedenlerinin gruplardaki rat sayılarının sınırlı olmasının bazen istatistiksel sonuçları sınırlandırabilmesi, çıkan anlamlı istatistiksel sonuçlarda da verilen tedavilerin etkinliğinin daha güçlü olmasının sonucu olduğu kanaatine varılmıştır.
Aims: In this study, it was aimed to experimentally test the contribution of traditional treatments with Platelet Rich Plasma (PRP) to hearing recovery in sudden hearing loss due to acoustic trauma. Matrials and Methods: In our study, 30 healthy adult rats were used. Before the study, otoacoustic emission (OAE) measurements of the experimental animals were made. All rats for which OAE measurements were made were exposed to 95 dB pure tone at 4 kHz for 60 minutes. It was determined that acoustic trauma occurred by measuring the otoacoustic emissions of the experimental animals at the 24th hour again. Of the rats divided into 3 groups of 10, the first group was the control group, the 2nd group was the group in which approximately 0.5 ml of intratympanic steroid was injected with 5 doses at 2 days apart, and the 3rd group was the group that was injected with approximately 0.5 ml of intratympanic PRP every 2 days. By evaluating the OAE measurements made approximately 10 days later, the treatments given were compared with the non-treated group and it was evaluated whether there was a statistically significant difference. Results: DPOAE measurements were compared on all days and the findings obtained from frequency measurements were compared statistically and it was checked whether significant results were obtained. It was determined that better results were obtained in those treated with PRP and prednol compared to rats with acoustic trauma who did not receive any treatment, and close treatment efficacy results were found between the PRP and prednol groups. Conclusion: In the comparison of DPOAE measurements of these three groups on four frequencies and on various days, there was a statistically significant difference in some days and frequencies, while in others, no significant difference was found. It has been concluded that the reasons for this are that the limited number of rats in the groups can sometimes limit the statistical results, and that the effectiveness of the treatments given is stronger in the significant statistical results.
