Özet
Bu tez çalışmasında Hatay ilinden toplanan Pleurotus ostreatus (Jacq. ex Fr.) P. Kumm., Agaricus arvensis Schaeff., Suillus luteus (L.) Roussel, Cantharellus cibarius Fr., Coprinus comatus (O. F. Müll.) Pers., Fomes fomentarius (L.) Fr., Trametes gibbosa (Pers.) Fr., Laetiporus sulphureus (Bull.) Murrill, Lycoperdon molle Pers., Cerrena unicolor (Bull.) Murrill mantarlarının Fenolik içerikleri, Element içerikleri, Toplam Antioksidan Seviyeleri (TAS), Toplam Oksidan Seviyeleri (TOS), Oksidatif Stres İndeksleri (OSI) ve Antimikrobiyal aktiviteleri belirlenmiştir. Mantarların TAS, TOS ve OSI değerleri Rel Assay kitleri kullanılarak belirlenmiştir. En yüksek TAS değerinin Coprinus comatus'da (5.225±0.086) olduğu görülmüştür. Ayrıca TOS değerleri incelendiğinde en yüksek TOS değerinin Cerrena unicolor'da (20.577±0.270) olduğu görülmüştür. Bunun yanı sıra OSI değerleri incelendiğinde en yüksek OSI değerinin Cerrena unicolor'da (2.151±0.085) olduğu belirlenmiştir. Mantar örneklerinden elde edilen etanol özütlerinin standart mikroorganizmalara karşı üremeyi engelleyici özüt konsantrasyonları agar dilüsyon metodu ile belirlenmiştir. Yaptığımız çalışmada kullanılan mantar özütlerinin mikroorganizmalara karşı farklı konsantrasyonlarda etkili olduğu gözlemlenmiştir. Ayrıca mantar örneklerinin bünyesinde bulunan bazı elementlerin (Cr, Cu, Mn, Fe, Ni, Cd, Pb, Zn) seviyeleri belirlenmiştir. Çalışmamızda mantarların bünyelerinde bulunan element düzeylerinin genel olarak civar illerde yapılan çalışmalardaki literatür aralıklarında olduğu görülmüştür. Fenolik içeriklerinin belirlenmesi amacıyla mantar özütleri SHIMADZU sistem HPLC cihazı ve DAD dedektör kullanılarak Caponio ve ark. (1999)'ne ait yöntem modifiye edilerek çalışılmıştır. Bu çalışma sonucunda elde edilen verilere göre mantarların bünyesinde acetohydroxamic acid, luteolin, catechin, vanilic acid, syringic acid, resveratrol, fumaric acid, gallic acid, caffeic acid, hydoxycinamic acid, hydroxybenzoic acid, salicylic acid, myricetin, ellagic acid, quercetin, butein ve naringenin varlığına rastlanmıştır.
In this thesis, Pleurotus ostreatus (Jacq. ex Fr.) P. Kumm., Agaricus arvensis Schaeff., Suillus luteus (L.) Roussel, Cantharellus cibarius Fr., Coprinus comatus (O. F. Müll.) Pers., Fomes fomentarius (L.) Fr., Trametes gibbosa (Pers.) Fr., Laetiporus sulphureus (Bull.) Murrill, Lycoperdon molle Pers., Cerrena unicolor (Bull.) Murrill mushrooms collected from Hatay province and phenolic contents, element contents, total antioxidant levels (TOS), total oxidant levels (TOS), oxidative stress indices (OSI) and antimicrobial activities of macrofungus were determined. TAS, TOS and OSI values of fungi were determined with using Rel Assay kits. The highest TAS value was observed in Coprinus comatus (5.225±0.086). In addition, when TOS values were examined, it was determined that the highest TOS value was in Cerrena unicolor (20.577±0.270). In addition, when the OSI values were examined, it was determined that the highest OSI value was in Cerrena unicolor (2.151±0.085). The concentrations of ethanol extracts obtained from mushroom samples against growth inhibition against standard microorganisms were determined by agar dilution method. It was observed that the mushroom extracts used in our study were effective against microorganisms at different concentrations. In addition, the levels of some elements (Cr, Cu, Mn, Fe, Ni, Cd, Pb, Zn) in the mushroom samples were determined. In our study, it was observed that the element levels in the fungi were generally within the range of the literature. In order to determine their phenolic content, mushroom extracts were analyzed by Caponio et al. (1999) was studied by modifying the method using SHIMADZU system HPLC device and DAD detector.
